The Biotrin Respiratory Viral Panel is an in vitro immnofluorescence assay for screening and identification of adenovirus, Influenza A, Influenza B, Parainfluenza 2, Parinfluenza 3 and respiratory syncytial virus (RSV) by indirect detection and in cell culture.
ASSAY PRINICPLE The Biotrin Respiratory Virus Panel utilises an indirect immunofluorecence antibody technique for identifying the 7 major viral respiratory pathogens in infected tissue cultures as well as in direct swab samples. The anti-viral screening reagent is used to confirm the presence of a respiratory virus. Specific virus identification is then achieved using the anti-adenovirus, anti-influenza A & B, anti-parainfluenza 1, 2 & 3 and anti-RSV monoclonal antibodies.
Fixed patient specimens are incubated with the anti-viral mouse monoclonal antibody on a glass slide. If specific viral antigen, is present a stable complex is formed with the antibody. Following a wash step, FITC labelled goat anti-mouse IgG is added which will bind to the antigen-antibody complex. Any unbound reagent is removed with a further wash step and the specimen is then visualised with the aid of a fluorescence microscope.
A positive reaction is denoted by bright green fluorescence. Uninfected cells appear dull red due to presence of Evan's blue counterstain in the FITC conjugate. In the Biotrin RVP kit it has been verified that the monoclonal antibodies directed against influenza A have positively detected the new strain of H1N1, most recently known as "swine flu".
